Climb Bio (NASDAQ: CLYM) lines up 2027 study after deep biomarker suppression

What happened

Climb Bio, Inc. (NASDAQ: CLYM) and Beijing Mabworks Biotech Co., Ltd. said on October 23, 2026, that they added more Phase 1 data for CLYM116/MIL116 in healthy volunteers. The anti-APRIL monoclonal antibody is being developed for IgA nephropathy. The studies in Australia and China were randomized, double-blind, placebo-controlled, ascending-dose trials. They enrolled 46 and 42 participants. A single subcutaneous dose of at least 480 mg kept free APRIL suppressed for 12 weeks.

A single 800 mg dose cut Gd-IgA1 by about 70% and IgA by about 60% at 12 weeks. The estimated half-life was about 33 days. The company said the antibody was generally well tolerated across the dose range. It presented the data at the American Society of Nephrology Kidney Week 2026 in Denver. NAVIGATE-2 is enrolling, and a global Phase 3 trial is expected in 2027, pending regulatory feedback.

Why it matters

OptimistFi's case is that Climb Bio needs clear clinical evidence before the balance sheet becomes the main issue. This filing supports that view. It shows durable biomarker effects, linear exposure, and no target-mediated drug disposition at doses of 160 mg or higher. That helps the case for a longer dosing interval, which matters for later-stage development in IgAN.

The safety data also look clean so far. There were no dose-limiting toxicities, treatment-related serious adverse events, or hypogammaglobulinemia. Injection-site reactions happened in 5 of 88 participants, or about 5.7%, and all were Grade 1 and went away without treatment. The downside is that these are healthy-volunteer data, not patient outcomes, and NAVIGATE-2 is still the test of the Q12W regimen.

What's next

NAVIGATE-2 initial data are expected in the first half of 2027. The company and Mabworks also expect to start a global Phase 3 trial in 2027, pending regulatory feedback. If patient data confirm the Q12W dosing regimen, they would support that Phase 3 plan. Weaker results or a delay would make the timeline less convincing.
